Properties of the Purified Hemolysin from a Psychrophile Isolated from Sea Mussel


Abstract
Hemolysin formed by a psychrophilic bacterium isolated from sea mussel was purified by ammoium sulfate precipitation, DEAE-cellulose column chromatography and sephadex G-100 column chromatography. The hemolysin was 100.7 unit/mg in specific
activity and
22.8 foldconcentrated than the original supernatant, and the yield was 0.3% at the final purification step. The purified hemolysin exhibited the optimum hemolytic activity at near 35¡É, but did not active at lower temperatures like below 5¡É.
Also
the
thermal stability of the isolated hemolysin was examined. The hemolysin kept 80% for the original hemolytic activity at near 20 ¡É and more residual activity than 95% was kept when treated at lower temperature than 10¡Éfor 10min. Neverthless, the
hemolytic substance was recognized as thermolabile hemolysin bcause lower activity than 5% for the original hemolytic activity was resided when treated at 35¡É.
The molecular weight of hemolysin was estimated at approximate 55,000 by using a sephadex G-100 gel filtration.
